Strain description

The B3gat1 KO mice are knockout mice deficient for galactosylgalactosylxylosylprotein 3-β-glucuronosyltransferase 1 (B3Gat1, aka., glucuronosyltransferase P, GlcAT-P), a synthetic enzyme of a neural specific carbohydrate antigen, the human natural killer-1 (HNK-1) carbohydrate antigen. The HNK-1 carbohydrate antigen is synthesized by a transfer of glucuronic acid to N-acetyllactosamine at non-reduced termini of carbohydrates by glucuronyltransferases: GlcAT-P (B3gat1) or GlcAT-S (B3gat2) and then a transfer of sulfate groups by natural killer-1 sulfotransferase (HNK-1ST). In the brain, GlcAT-P (B3gat1) is a major glucuronyltransferase involved in the HNK-1 carbohydrate antigen synthesis. The B3gat1 KO mice exhibited normal development of gross anatomical features, but the adult mutant mice exhibited reduced long-term potentiation at the Schaffer collateral-CA1 synapses and a defect in spatial memory formation. The strain is useful as a mouse model for analyzing high-order functions of the brain.
