The oma mice are skeletal anomaly mutant found in 4 way cross strain derived from C57BL/6J, C3H, DBA/2 and BALB/c maintained in School of Agriculture, Okayama University. These mice observe remarkable deformity and fusion of vertebrae, and deformity and bifurcation of ribs. It shows that phenotype of oma mice causes to change 409th glycine residue to cysteine by single base substitution occurred on 8th exon of Delta-like3 (Dll3) gene. C57BL/6-Dll3oma is a congenic strain by 17 times backcrossing to C57BL/6J with original oma mice.
Genotyping of Dll3oma is able to carry out following PCR-RFLP. Folloing two primer is used for PCR. Forward primer was designed as mismatch primer by substituting CAGC at the 3'-terminal for CGGC derived from original genome in order that transgene is digested by PvuII.
PCR carry out following conditions.
Two genotypes are discriminated by agarose gel electrophoresis after PCR productions are digested by restriction enzyme PvuII.
