Strain description | Three mRNA isoforms (PGC-1α-a, -b, and -c) are transcribed from PGC-1α (Ppargc1a) gene. Slightly different proteins are translated from the three mRNA isoforms. Expression of each isoform is different depending on tissue types and stimuli. Exercise increases PGC-1α-b and PGC-1α-c mRNAs, but not PGC-1α-a mRNA, in the skeletal muscle. On the other hand, fasting increased PGC-1α-a mRNA, but not PGC-1α-b or PGC-1α-c mRNAs, in the liver. The PGC-1α-b/B-line is a transgenic mouse line with skeletal-muscle-specific (driven by human α-actin promotor) overexpression of PGC-1α-b mRNA (derived from the line 03-2 in Table 1 of Ref. 1). This strain is useful for studying isoform-specific functions of PGC-1&alpha mRNAs and proteins in skeletal muscles. Multiple sublines overexpressing each isoform in skeletal muscles are available: three sublines (nbio095, nbio096, and nbio097) for PGC-1α-a, two sublines (nbio299, and nbio300) for PGC-1α-b, and three sublines (nbio301, nbio302, and nbio303) for PGC-1α-c. |
